Is it a must that there should be internet connection to play Pokemon go?

Internet connection means wifi or mobile data.

In theory, no, but it would require Niantic to re-code the game to accept accelerometer data from your phone to replace constant GPS updates.  Even then, it would still need to double check with GPS periodically.

Unfortunately, yeah. Without an internet connection a GPS signal can't be found. It's kind of a drag for poor people like myself lol
